Aculeus uses pretty much the same setup we do for Broadcasting Live Dota Matches, except I connect to my co-broadcaster with Skype and broadcast audio directly on the Video Stream!

Ok, Stayed up till 5am working this crap, but Finally got a good version of FFDShow that worked, had to use the Manual command line encoder and now it all works fine :D

You're gona need a 50meg upstream to handle that many people at that bitrate. So you're gona be looking at a 100meg up/down fiberoptic connection server. Which ISNT cheap by any sense of the word.
